PCR conditions :

Standard PCR buffer conditions and Taq-DNA polymerase

First step 2 min at 94°C
30 cycles 40 sec at 94°C
45 sec at 61°C
1 min at 73°C
Final step 4 min at 73°C

Expected PCR products :

• wild type allele oligo(+/-) and oligo (+) combination 200 base pairs
• mutated allele oligo(+/-) and oligo (-) combination 700 base pairs
